Chloe Sullivan and Lois Lane are cousins who are as close as sisters. Lois is at least a year older than Chloe. Chloe spent her childhood in Metropolis and her teenage years in Smallville, while Lois spent her childhood traveling around the world with her military father Sam Lane. However, the two were still able to cultivate a close relationship across the distance. Chloe first mentioned her cousin's name when she attempted to write under a pseudonym after being blacklisted by Lionel Luthor.

Since Lois moved to Smallville in 2004, their relationship has grown even closer. Lois found friendships in Chloe's close friends and a surrogate family in the form of Chloe's best friend, Clark Kent, and his parents. They help each other through crises, defend each other fiercely, and don't hesitate to risk their lives for the other. Chloe and Lois often rely on each other for moral and family support such as taking each other to the hospital or turning to the other for romantic relationship advice. When the Metropolis University dorms closed in 2006, Lois offered Chloe her apartment at the Talon, which they continued to share until Chloe became engaged. Both girls have an insatiable curiosity and often worked together to get to the bottom of the strange occurrences of Chloe's home town. As they have gotten older, they were simultaneously trying to cultivate successful journalism careers, forging an initial rivalry, but settling into supporting each other. However, while Lois has continued her path as a journalist, Chloe has attempted to try different career paths.

Because she cares deeply for both Lois and Clark, Chloe delicately balances both relationships. Constantly trying to protect Clark's secret often directly conflicts with her support of her cousin. At the same time, Lois has been unknowingly saved by Clark several times; she almost always calls Chloe first when she is in danger, and Chloe inevitably sends Clark to rescue her.

Curiously, despite their close relationship, Chloe doesn't always confide in Lois about her greatest insecurities. She has refused to tell Lois about her mother's mental illness and her own meteor rock infection. Chloe tries to protect Lois from making hasty, foolish decisions. Alternately, Lois is keenly aware of how much Chloe cares for Clark and tries to make sure Chloe doesn't sacrifice too much of herself for the sake of others.

Lois' arrival in Smallville was the direct result of an elaborate scheme to protect Chloe from Lionel Luthor. He commissioned to have Chloe and her father killed after she testified against him and sent him to jail. Chloe and Gabe were kept in hiding by Lex Luthor and Lois' own father General Sam Lane. Lane's involvement in Chloe's faked death was probably how Lois was able to obtain a video letter Chloe made for Clark Kent, pleading with him to get to the bottom of Lionel's machinations. Lois went to Smallville to find Clark and question how he knew her cousin. She even dug up Chloe's grave after Clark suggested she wasn't really dead, and the two worked together to locate Chloe before another attempt was made on her life. After finding her cousin safe and sound, Lois intended to leave Smallville for her first semester at Metropolis University, but was informed that she hadn't passed her senior year. Her father ordered her to obtain her remaining credits at Smallville High. Chloe was thrilled to attend school with Lois and encouraged her to get involved by writing for her paper, the Smallville Torch. Lois was initially reluctant but her first effort was well-received. While staying with the Kents, Lois realized her cousin was deeply in love with Clark, but was outraged that he only wanted to be platonic friends. She often offered her opinion on their complicated relationship with the intention of protecting Chloe from heartbreak. Chloe introduced Lois to all of her close friends and frequently got her older cousin involved in the bizarre events that occur in the town. Lois threw her a wild eighteenth birthday party and even attended the prom to see Chloe crowned Prom Queen. When the second meteor shower struck the town in May, Chloe and Lois tried to evacuate together, but Chloe instructed Lois to leave without her so she could check on Lana.

Chloe moved to Metropolis to attend Metropolis University in the fall of 2005, while Lois continued to live with the Kents. Chloe also began interning at the Daily Planet, answering the 24-hour news hotline, and Lois would often keep her company. She and Lois investigated the disappearance of dancers at the Windgate Club by pretending Lois was auditioning for a role and made her perform a sexy dance so she can have time to interrogate the other exotic dancers. Lois was promoted as Jonathan Kent's campaign manager and asked Chloe to help her dig up dirt on his opponent, Lex Luthor. Chloe also helped Lois move out of the Kents' and into the Talon apartment. There, Chloe was possessed by the spirit of a murdered girl and seemingly attempted suicide. A worried Lois took her to the hospital and blamed herself for not realizing Chloe was in danger of harming herself. They wound up being kidnapped by the girl's murderer, Michael Westmore, but were able to save themselves. Despite Lois' obvious concern, Chloe chose not to tell Lois that her mother was in a mental institution. Chloe and Lois attended the funeral of Jonathan Kent together and tried to help Martha and Clark cope. They helped Martha throw Clark a surprise birthday party to cheer him up.

During Chloe's second year in college, she moved into Talon apartment with her cousin and slept on a futon. Lois decided that journalism was also her calling and began to investigate the Kents' errant barn door. Trying to divert Lois' attention from Clark's superpowers, Chloe discouraged her investigation and was not supportive of Lois' first effort. Undeterred, Lois shopped her story to the Metropolis Inquisitor and landed a job reporting for the tabloid. Chloe did help Lois investigate the secret identity of the Green Arrow, but abruptly stopped when Clark asked her not to. Lois called Chloe for help when she was kidnapped by enemies of Green Arrow and she and Clark were able to save her. Chloe and Lois attended the Kents' Thanksgiving Day celebration. Even though she figured out Oliver Queen's secret identity, she encouraged Lois' relationship with him, helping her pack for a romantic getaway. Lois, in turn, was supportive of Chloe's relationship with Jimmy Olsen. Jimmy thought Lois and Clark might make a good match and set up a blind date for the two on Valentine's Day. Affected by a love-potion, Lois became infatuated with Clark. Chloe was amused by her actions until Clark seemingly returned the affection, alerting Chloe (and Martha) that Clark and Lois were infected with red kryptonite. Chloe and Jimmy cured them and Lois did not remember the incident. Lois heard Clark and Chloe talking about an underground fight club and stole their notes in order to investigate the story. As Lois and Chloe got closer to learning the full extent of Lex Luthor's latest Level 33.1 project, Project Ares, Lois became the target for an experimental assassin. Clark and Chloe worked together to find her and save her. Chloe pleaded with Lois to not put herself in danger again by snooping further. Fueled by Lex's victimization of her childhood friend Wes Keenan, Lois ignored Chloe's warning and tracked down Lex's lab at Reeves Dam. She was stabbed in the stomach by a guard and called Chloe for help. Chloe found her cousin near death from blood loss. Devastated, Chloe's latent meteor power manifested and healed Lois, but rendered Chloe herself unconscious.

After Clark saved both girls from the ruins of the dam, he and Lois waited anxiously for Chloe to be revived. Lois was unable to just sit and wait, and asked Clark to stay so she could go look for clues. Much to Clark's devestation, Chloe was pronounced dead by medical staff, but inexplicably revived shortly after. She refused to tell Lois what happened, burning the evidence of her death certificate. Lois continued to investigate Reeves Dam and discovered a spaceship. She wrote a story about it and showed it to Chloe. Chloe recognized it as Kryptonian, and once again trying to maintain secrecy, said the story was not Daily Planet material. However, Grant Gabriel, Chloe's new managing editor, immediately took a liking to Lois and challenged her to develop the story. Lois tried to tell Grant that she would not betray her cousin, but Grant appealed to her ambition and tenacity, and Lois was unable to resist. He indeed hired Lois and gave her a desk right next to Chloe.

Chloe waited with Lois at the hospital after she was knocked unconscious by a super-powered Lana Lang.

Grant frequently commented that Lois' talent and competition could spark Chloe's lost passion again, but Chloe and Lois remained on good terms, even helping each other out with their stories. When Lois was picked to write an exposé on Lex Luthor, Chloe gave her a list of questions to ask him. Grant proved to be extremely divisive between the cousins when he and Lois began a relationship. Chloe knew Lois was hired because of her potential merit, but she was worried that her reputation would be ruined. She gave Lois a stern lecture urging her to break it off with Grant. Lois acknowledged that it was a poor decision, but continued to see Grant without Chloe's knowledge. Lois and Chloe's relationship was used against them by Adrian Cross. Wanting to expose Lex's wrongdoings, he threatened Chloe's life to manipulate Lois into following his orders. Lois tried desperately to save Chloe, but in the end, she was saved by Clark's evil doppelganger, Bizarro. After Lois followed Lex to Detroit and located Kara, Chloe teamed up with Clark to rescue them from Finley, who shot Lex, then kidnapped them. Chloe had to use her powers to revive Lex, but was dead for a considerably longer length of time. Upon waking, one of Chloe's first concerns was if Lois had seen her dead, but Clark assured her that she hadn't.

After Chloe's arrest and detainment, Lois correctly deduced that Lex Luthor had been involved and broke into the Luthor mansion in an attempt to find information. She discovered Chloe's location and went undercover to rescue her.

Lois moved out of the Talon apartment when Chloe and Jimmy got engaged. She got drunk at their engagement party, admitting that she didn't really think Chloe and Jimmy were meant to be. Afterward, she was very embarrassed and apologetic, explaining that she didn't want Chloe to get hurt like she did with Oliver and Clark did with Lana. She gave Jimmy an awkward hug and welcomed him into the family.

Lois was in charge of planning of Chloe and Jimmy's wedding as the maid of honor. On the big day, she asked Chloe how she knew Jimmy was the one. When Jimmy was severely injured and Chloe went missing, Lois accompanied him to Star City in Chloe's place. When Lana Lang returned to Smallville, Clark and Lana rekindled their relationship. Knowing that Lois' feelings towards Clark had changed, and Clark was considering his feelings towards Lois as well, Chloe directly warned Clark against hurting Lois while he tried to figure out what he wanted.

During Chloe's birthday, she admitted jealousy of Lois' life and then wished she could have it. A magician took her wish literally and Chloe woke up in Lois' body. She lived a day in Lois' life as a reporter, believing that Lois was popular at the Daily Planet and that Clark gave Lois "puppy-dog eyes", and told Lois that "Clark doesn't mind her assertive flair." After the switch back, Chloe decided her dream of reporting was living in the past and that she wanted to try something new.

Chloe came to pick up Lois after she was not allowed into a convention and reassured her after she was doubting herself as a reporter and the avoidance of the Red Blue Blur. While Chloe got mugged, Lois fought off her attackers and one escaped with Chloe's car and laptop. Lois created "Stiletto" and pretended to be a super hero in order to meet the Red Blue Blur and to jumpstart her career again. Later, Lois confessed that she was in over her head and trying to correct the fiasco. Chloe was prompted to almost confess her involvement with Davis Bloome, but decided not to.

After Chloe left town with Davis Bloome to protect Clark, Lois believed Clark should have done more to find her. Clark later called her as the Red Blue Blur and promised he was looking for Chloe. Directly after, Lois went missing, after accidentally picking up a magical time-traveling ring. Clark then told Chloe that he'd looked everywhere for her, to no avail.

After missing for three weeks, Lois mysteriously returns from the future, with no memory of ever being there. Upon her arrival, Lois ends up on a Metropolis monorail and finds herself under attack by a female assassin, who was also transported back from the future. While the pair fight, the train loses control and derails off the track. After the crash, Lois ends up at hospital and reunites with Chloe there.
